This kit is intended for scientific research only and not for medical diagnosis. It is designed for the quantitative detection of Human Glycophosphatidylinositol (GPI) in serum, plasma, and related fluid samples.
**Principle of Detection:**
The kit uses a two-antibody sandwich ELISA method. After incubation, unbound components are washed away. A substrate (TMB) is added, which changes color under the action of horseradish peroxidase (HRP). The intensity of the color is directly proportional to the concentration of GPI in the sample. The optical density (OD) is measured at 450 nm, and the concentration is calculated based on a standard curve.

- 4. 20× concentrated washing solution (20 mL)

**Preparation Instructions:**
Dilute the 20× washing solution with distilled water to the working concentration. Prepare standards by diluting the stock solution to concentrations of 200, 100, 50, 25, 12.5, and 6.25 ng/mL.
**Required Equipment:**
- Incubator (37°C)
- Microplate reader
- Pipettes and disposable tips
- Distilled water
- Disposable test tubes
- Absorbent paper
**Procedure Summary:**
1. Bring the kit to room temperature (30 minutes).
2. Dilute the washing solution.
3. Add standards and samples to the plate.
4. Incubate at 37°C for 30 minutes.
5. Wash the plate 4 times.
6. Add enzyme working solution.
7. Incubate again for 30 minutes.
8. Wash the plate again.
9. Add substrate A and B, develop color in the dark for 15 minutes.
10. Stop the reaction with stop solution.
11. Measure OD values at 450 nm within 15 minutes.
12. Calculate the sample concentration using the standard curve.
**Sample Requirements:**
- Avoid sodium azide (NaN₃), as it inhibits HRP.
- Store samples at -20°C if not tested immediately.
- Centrifuge samples to remove particles and hemolysis.
**Precautions:**
- Follow the manual strictly.
- Seal unused enzyme labels with desiccants.
- Perform duplicate tests for accuracy.
- Note the dilution factor when calculating results.
- Ensure all measurements fall within the detection range (0.1–200 ng/mL).
- Avoid cross-contamination and direct light exposure for substrate B.
**Storage & Shelf Life:**
Store at 2–8°C, away from light and moisture. Shelf life is 6 months.
