After washing, the samples are divided into two groups: one group contains both the tested antigen and the enzyme-labeled antigen, while the other group only contains the enzyme-labeled antigen. Following incubation and substrate addition, a color reaction occurs. By comparing the intensity of the color between the experimental and control groups, we can determine the concentration of the unknown antigen in the sample.
This technique is widely used in ELISA assays for small molecules like hormones and drugs, where traditional sandwich methods may not be applicable.
Brief Steps of the Competitive Method:
- 1) First, an antibody is immobilized onto the surface of the microplate.
- 2) A mixture of the sample and enzyme-labeled antigen is added, along with a control group containing only the enzyme-labeled antigen.
- 3) After incubation, the substrate is added, triggering a color reaction. The difference in color intensity between the experimental and control groups is used to calculate the amount of the unknown antigen.
